Four Strep-tagged proteins were purified using StrepCaptureXP™ Resin Lite (GenScript, L01114) and a commercial affinity resin from Competitor-ST under identical purification conditions. Elution fractions were analyzed by SDS-PAGE.

The target membrane protein (ASIC2a) has a theoretical molecular weight of 60.1 kDa.
Sample Preparation:
1. Cells were lysed in 20 mM HEPES, 150 mM NaCl (pH 7.4) containing 1–1.5% (w/v) LMNG for 2 h at 2–8°C.
2. The lysate was clarified by ultracentrifugation at 45,000 rpm for 45 min at 4°C.
3. 100 μL of resin was incubated with 12 mL of clarified membrane protein lysate for 2 h at 2–8°C.
4. After binding, washing and elution were performed according to the recommended purification protocol. Elution fractions were analyzed by SDS-PAGE. »
Binding capacity of StrepCaptureXP™ Resin Lite (GenScript, L01114) was evaluated after repeated regeneration with 0.1 M NaOH. No significant loss in binding capacity was observed after 40 regeneration cycles. »
