Custom Vector Construction A panel of lysine-to-alanine PRDM9 singlemutant plasmids (K207A, K226A, K297A, K338A, K368A, K372A, K374A and K375A) were generated through synthesis of DNA-coding sequences followed by insertion into the pET21a(+) expression vector via the NdeI and BamHI insertion sites (GenScript).Inhibition assays of PRDM9 automethylation were performed by pre-incubating 5 mM PRDM9 with varying concentrations of suramin (Sigma–Aldrich), sinefungin (Sigma–Aldrich) or H3pNle (synthesised by GenScript) for 30 min at room temperature before the addition of SAM.
