T cell receptor (TCR) based immunotherapy is an attractive strategy to target a wide range of intra-tumoral antigens and elicit robust tumor cytotoxicity. However, engineering soluble TCR engagers that preserve physiological affinity is crucial for universal TCR drug development, yet remains challenging.
