Gene Synthesis Thrombin and adenosine were purchased from Sigma-Aldrich. TMPyP4 (tetra-(N-methyl-4-pyridyl) porphyrin) tosylate was obtained from Merck. The oligonucleotides used here were synthesized and purified by Nanjing Genscript Co. Ltd. (Nanjing, China). The acceptor bead (Catalog number 6772001) and the donor bead (Catalog number 6760002S) were obtained from PerkinElmer BioSignal, Inc. Other chemicals were of analytical grade. All aqueous solutions were prepared using double-distilled water (ddH2O), which was purified with a Milli-Q purificationsystem (Branstead) to a specific resistance of >18 MΩ. Get A Quote

摘要

A major challenge in the field of biosensing is high-throughput screening (HTC) of small molecules in complex biological media such as human serum. Two aspects are required to achieve the goal: (1) General assays for target-independent detection of different molecules specifically and sensitively; (2) the operation is able to be standardized with automation possibility. This work integrates the distinguishing features of aptamer and fluorescent techniques. Innovatively, a very easy method is designed to combine a long range diffusion triggered excitation and a great steric hindrance effect, which makes it possible to become a general sensing method. As a proof-of-principle, adenosine in undiluted serum was dete... More
