Peptide Synthesis> | 3D PEG-maleimide (PEG-Mal) hydrogels were prepared at 20 wt% solution with a 20K 4-arm PEG-Mal (Jenkem Technology, Plano, TX) crosslinked at a 1:1 ratio with 1.5K linear PEG-dithiol (Jenkem) in 2mM triethanolamine (pH ~ 7.4). Cells were pelleted and resuspended in PEG-Mal solution before mixing with PEG-dithiol. The cell adhesion peptides (GenScript, Piscataway, NJ) CGP(GPP)5GFOGER(GPP)5 (1mM), CGPHSRN(G)6RGDS (0.8mM), CGGSVVYGLR (0.1mM), and CGGAEIDGIEL (0.1mM) were reacted with PEG-Mal 10 minutes before gelation.
