F-Luc Self-Amplifying RNA (Cap1, m5C)

The F-Luc self-amplifying RNA (saRNA) expresses firefly luciferase protein, which is the enzyme for bioluminescence of fireflies and click beetles. This saRNA is capped with Cap1-AU structure with high capping efficiency. It has 100% substituted with 5-methylcytidine for enhanced expression and reduced immunogenicity. The GenScript patented saRNA vector 1 is used. The saRNA has a 68A tail in its sequence. After efficient delivery, F-Luc saRNA can translate into F-Luc and emit bioluminescence with Luciferin.

Description

The Firefly-Luciferase (F-Luc) self-amplifying RNA (saRNA) expresses firefly luciferase protein, which is the enzyme for bioluminescence of fireflies and click beetles. After efficient delivery into cells or animals, F-Luc mRNA can translate into F-Luc and emit bioluminescence in the presence of its substrate, Luciferin.  
 
This saRNA features: 
- A Cap1-AU structure with high capping efficiency 
- 100% substitution with 5-methylcytidine (m5C) for improved protein expression and reduced innate immune response 
- Constructed using GenScript’s patented saRNA Vector 1 
- A 68A poly(A) tail to mimic natural mature mRNA

Transfect 0.01-0.05 µg of saRNA with 0.5 µL of Lipofectamine™ 2000 (or equivalent reagent) per well in a 96-well plate, following the manufacturer’s protocol. Assess luciferase expression 12-48 hours post-transfection using luciferase assay system (Promega, E4030).
