Gene Synthesis A 118-nucleotide sequence encoding SmallTalk-Bin1b (Bin1b accession number in Antimicrobial Peptide Databases = AP01592) optimized according to the codon usage preferences in E. coli was synthesized by GenScript (Piscataway, NJ, USA) and cloned into pUC57. For antimicrobial assays comparison with SmallTalk-Bin1b, the SmbP-Bin1b DNA construct was synthesized and cloned into pET-30a(+) by GenScript. Recombinant protein production in Escherichia coli is a fundamental aspect of biotechnology. Fusion tags are commonly used to enhance solubility and facilitate purification. However, these tags can lead to challenges such as low yields, complicated purification processes, and the necessity for tag removal, especially when dealing with peptides. This study introduces a novel fusion tag called SmallTalk, a truncated version of the small metal-binding protein SmbP.
