Description

The GenLNP-S01 Negative Control contains lipid nanoparticles (LNPs) encapsulating a non-coding actin RNA, designed to serve as a biologically inert control in RNA delivery studies. The LNP is formulated with SM102 as the main ionizable lipid for efficient encapsulation and cellular delivery.  
 
This LNP features: 
- Non-coding actin RNA engineered by removing the ATG start codon and downstream initiation signals to prevent protein translation 
- Cap1 structure for co-transcriptional capping and a poly(A) tail to mimic mature mRNA stability 
- Maintains native RNA sequence composition to minimize nonspecific cellular responses 
- Biologically inert design providing high biocompatibility and reliable baseline control 
 
This Negative Control LNP provides a stringent reference for evaluating delivery efficiency and cytotoxicity, enabling clear and reproducible interpretation of experimental results.

1. LNP can be added directly to cells incubated in complete media  
2. Cells grown and transfected in complete media (DMEM +10% FBS) has higher expression than reduced serum medium  
3. For suspension cell lines, we recommend using low fluorescent background media such as Flourobrite® with 10% FBS, 1X GlutaMax® and 1X sodium pyruvate  
4. For adherent cell line, we recommend replace complete media with Opti-mem right before reading the cell eGFP expression  
5. For higher expression, we recommend incubate the LNP with FBS at 1:2 volume ratio (LNP:FBS) for 30 minutes at 37C before transfecting
