The Cas13b mRNA encodes Cas13b, an RNA-guided ribonuclease from the type VI CRISPR-Cas system. Cas13b mediates targeted cleavage of single-stranded RNA through complementary binding of a CRISPR RNA (crRNA) spacer to its RNA target, enabling RNA knockdown and transcriptome engineering applications. | A549 cells were seeded in a 96-well plate at 10,000 cells per well. After overnight incubation, the cells were transfected using Lipofectamine™ MessengerMAX™ (or an equivalent reagent) with 300 ng of Cas13b mRNA, 100 ng of crRNA, and 100 ng of Gluc mRNA per well. The cells were incubated for 24 and 72 hours post-transfection. Three crRNA sequences and one non-targeting control (NTC) were tested. |
