eGFP mRNA (Cap1, m1Ψ)

The eGFP mRNA expresses an enhanced green fluorescent protein (Ex: 488nm, Em: 507nm). This mRNA is capped with Cap1 structure with high capping efficiency. It has 100% substituted with N1-methyl-pseudo Uridine for enhanced expression and reduced immunogenicity. The GenScript patented UTR is used in this mRNA. The mRNA has a 100A tail in its sequence, mimics a mature mRNA. EGFP mRNA is a commonly used fluorescence reporter sequence in mRNA therapeutics and delivery system development projects.

Description

The eGFP mRNA encodes an enhanced green fluorescent protein, originally derived from the jellyfish, Aequorea victoria. eGFP mRNA is widely used as a fluorescent reporter in mRNA-based research, especially in therapeutic development and delivery system validation. The expressed eGFP protein fluoresces green, with excitation at 488 nm and emission at 507 nm.  
 
This mRNA features: 
- A Cap 1 structure with high capping efficiency 
- 100% substitution with N1-methyl-pseudouridine (m1Ψ) for improved protein expression and reduced innate immune response 
- GenScript’s patented UTR to enhance translation 
- A 100A poly(A) tail to mimic natural mature mRNA 
 
This construct offers strong, reliable fluorescent signaling, making it ideal for transfection optimization, biodistribution studies, and LNP delivery validation.

Transfect 0.1-0.2 µg of mRNA with 0.5 µL of Lipofectamine™ 2000 (or equivalent reagent) per well in a 96-well plate, following the manufacturer’s protocol. Analyze the expression 16-48 hours post-transfection using a plate reader, flow cytometer, fluorescence microscope, or confocal microscope. For equipment settings, we recommend: 485 nm excitation, 535 nm emission.
