Recombinant HEK293 cells stably contain a luciferase reporter gene under the control of SIE responsive element (SIE-RE). IL-6 can activate the receptor on the cell surface and transmit the signal to the downstream signaling pathways. The luciferase will be translated when the receptor was activated. This stable cell line product is designed for In Vitro cell-based functional assays to determine the bioactivity of IL-6.

Culture Properties Adherent
Stability Stable through more than 20 passages with no significant changes in assay performance or expression profile.
Size Two vials of frozen cells (>1×106 per vial in 1 mL).
Storage Store cells in liquid nitrogen immediately upon receipt. Thaw and recover cells within one year from the date received.

Figure 1. IL-6-induced luciferase expression in HEK293/Luc (IL-6) cells. After stimulation by agonist IL-6 (Cat. No. Z03034, Genscript) for 6 hours, the cells are determined with Fire-Lumi™ Luciferase Assay System (Cat. No. L00877C-100, Genscript) and the relative luminescence units (RLU) were recorded by plate reader (Pherastar, BMG). The RLU were plotted against the log of the cumulative doses of IL-6 (Mean ± SEM, n = 3). The EC50 of IL-6 on this cell was 35.34 ng/ml.

Notes:
EC50 value is calculated with four parameter logistic equation:
Y=Bottom + (Top-Bottom) / (1+10^((LogEC50-X)*Hill Slope))
X is the logarithm of concentration. Y is the response.
Y is RLU and starts at Bottom and goes to Top along a sigmoid curve. »
