Figure 1. EGF-induced luciferase expression in HEK293/SRE-Luc cells. After stimulation by EGF (Z02691-100, Genscript), the cells are determined with Fire-Lumi™ Luciferase Assay System (Cat. No. L00877C-100, Genscript) and the relative luminescence units (RLU) were recorded by plate reader (Varioskan LUX, Thermo Fisher). The RLU were plotted against the log of the cumulative doses of EGF (Mean ± SEM, n = 3). The EC50 of EGF on cells was 1.84 ng/ml.

EC50 value is calculated with four parameter logistic equation:
Y=Bottom + (Top-Bottom) / (1+10^((LogEC50-X)*Hill Slope))
X is the logarithm of concentration. Y is the response.
Y is RLU and starts at Bottom and goes to Top along a sigmoid curve.

Figure 2. Functional evaluation of Cetuximab, the EGFR antibody, in HEK293/SRE-Luc cells. HEK293/SRE-Luc cells were dispensed into the microplate and incubated at 37℃ overnight prior to the addition of different concentrations of Cetuximab. After 30 minutes’ incubation, EGF was added into the plate and incubated at 37℃ for 6 hours. Luminescence signal was detected with the plate reader (Varioskan LUX, Thermo Fisher). »
