| Product Description |
The GenBuilder™ Ultra Cloning Kit enables seamless and one-step cloning of multiple DNA fragments into a linearized vector. The proprietary enzyme mix fuses together DNA fragments efficiently and precisely by recognizing 15-40 bp overlaps at the end of each fragment. The system enables seamless cloning of DNA fragments into any linearized vector, requires no pre-existing recombination sites or helper sequences, and eliminates the need for traditional restriction digestion and ligation workflows. The GenBuilder™ Ultra Cloning kit can simultaneously clone up to 10 DNA fragments into a linearized vector. The kit can also be used to assemble single stranded DNA (ssDNA) oligos with double stranded DNA (dsDNA) fragments, and using ssDNA as a linker to fuse together two adjacent dsDNA fragments. |
| Storage |
The kit contains GenBuilder™ Ultra DNA 2× Assembly Mix and Positive Control. Keep all components at –20°C for long term storage. Thaw the master mix on ice prior to use. For the best performance, please aliquot the master mix into smaller volumes and avoid repeated freeze-thaw cycles. |
Three, five, and ten fragments (each fragment at ~0.025 pmol, with 30-bp overlaps) were assembled using the GenBuilder™ Ultra Cloning Kit to construct the pUC57 vector. Assembly reactions were performed at 50°C for 40 min. 2 μl of assembled mix was transformed into Top10 Competent E. coli and spread on LB/Amp plates. »
Ten DNA fragments (~0.025 pmol each, with 30-bp overlaps) were assembled using the GenBuilder™ Ultra Cloning Kit to construct pUC57, pFastBac™ (a trademark of Thermo Fisher Scientific), and pET vectors. Assembly reactions were performed at 50°C for 40 min. 2 μl of each assembled mix was transformed into Top10 Competent E. coli and spread on LB/Amp plates. Data suggests GenBuilder Ultra is compatible with a variety of vectors. »
Four different fragments (~0.025 pmol, ~3.3 kb each) with 30 bp overlap were assembled using GenBuilder™ Ultra Cloning Kit into a 6.0 kb vector. Assembly reactions were performed at 50°C for 40 min. 5 μl of each assembled mix was transformed into Top10 Competent E. coli and spread on LB/ Amp plates that indicated assembly were analyzed with PCR. Each fragment adapter (AB, BC, CD) was verified with DNA gels, indicating that the fragments were successfully assembled in the correct order into the vector. »
