CHO-K1/CASR Stable Cell Line

Figure 1. CaCl2-induced concentration-dependent stimulation of intracellular calcium mobilization in CHO-K1/CASR cells. The cells were loaded with Calcium-4 prior to being stimulated with agonist CaCl2. The intracellular calcium change was measured by FLIPR. The relative fluorescent units (RFU) were normalized and plotted against the log of the cumulative doses (5-fold dilution) of CaCl2 (Mean ± SD, n = 2). The EC50 of CaCl2 on this cell was 0.43 mM.
Notes:
1. EC50 value is calculated with four parameter logistic equation:
Y=Bottom + (Top-Bottom)/(1+10^((LogEC50-X)*HillSlope))
X is the logarithm of concentration. Y is the response
Y is RFU and starts at Bottom and goes to Top with a sigmoid shape.
2. Signal to background Ratio (S/B) = Top/Bottom

CHO-K1/CASR Stable Cell Line

CASR is a calcium-sensing receptor and plays an important role in regulating PTH secretion. It is expressed in many different tissues, such as parathyroid, pancreatic islet and insulinoma cells, breast, gastrinoma cells, pituitary, keratinocytes and human colon epithelial cells. CASR Inhibits PTH secretion and parathyroid cellular proliferation in parathyroid glands or cells, and also inhibits proliferation of colonic crypt cells. Besides, it can stimulate PTHrP secretion in prostate cell line, and stimulate calcitonin secretion from thyroidal C-cells.
