1.酵母系统表达蛋白
金斯瑞YeastHIGHTM 表达系统以及YeastFast技术在分泌表达方面具有很强的优势。下图所示为使用金斯瑞YeastHIGHTM 表达技术表达一种酶。SP-Sepharose一步法纯化后,产生了表达水平约20mg/L的无标签蛋白,且纯度高于90%。金斯瑞将通过MALDI-TOF方法进一步检测目标蛋白。

2.信号肽
下图所示为一种信号肽蛋白酶的表达情况。使用Saccharomyces cerevisiaeα factor prepro peptide(α因子前体肽原)表达,无法表达目的蛋白。然而,将α因子前体肽原序列更换为蛋白酶信号肽自身序列时,酵母成功分泌了目的蛋白,目的蛋白正确表达。
